Differing Responses to Cryphonectria parasitica at Two Indiana Locations

چکیده
Chestnut blight, a disease that has spread rampantly among American (Castanea dentata (Marsh.) Borkh.) and European chestnut (C. sativa Mill.) trees, results from infection by the fungal pathogen Cryphonectria parasitica (Murrill) M.E. Barr parasitica). This fungus was introduced in early 1900s almost functionally eliminated trees North landscape. In 2017, we collected blight samples two sites (Site B, (Fulton Co., IN) Site C (Marshall IN)). At Fulton County planting, cankers had formed, healed over, were healthy. However, at second site Marshall County, C), continued to propagate until all of died back ground. Research evidence worldwide indicated these visual clues likely result presence hypovirus. Upon closer inspection subsequent isolation reproduction spores, no hypovirus been identified either site. Here, present curious coincidence where one completely succumbed disease, while other able spring health.

Mycovirus cryphonectria hypovirus 1 elements cofractionate with trans-Golgi network membranes of the fungal host Cryphonectria parasitica.
The mycovirus cryphonectria hypovirus 1 (CHV1) causes proliferation of vesicles in its host, Cryphonectria parasitica, the causal agent of chestnut blight. These vesicles have previously been shown to contain both CHV1 genomic double-stranded RNA (dsRNA) and RNA polymerase activity. To determine the cellular origins of these virus-induced membrane structures, we compared the fractionation of se...
متن کاملAlternating Search at Two Locations
In this paper we introduce a new class of search problem which we calìalternating search'. Two searchers starting at given points in diierent search regions, who can move alternately with speed one, or (as a limiting case) simultaneously with combined speed one, seek to nd (reach) an object in least expected time. The hidden object is stationary and its location is given by a known distribution...
متن کاملMultilocus PCR Assays Elucidate Vegetative Incompatibility Gene Profiles of Cryphonectria parasitica in the United States.
Chestnut blight is a devastating disease of Castanea spp. Mycoviruses that reduce virulence (hypovirulence) of the causative agent, Cryphonectria parasitica, can be used to manage chestnut blight. However, vegetative incompatibility (vic) barriers that restrict anastomosis-mediated virus transmission hamper hypovirulence efficacy. In order to effectively determine the vegetative incompatibility...
متن کاملHeterologous expression of a tannic acid-inducible laccase3 of Cryphonectria parasitica in Saccharomyces cerevisiae
BACKGROUND A tannic acid-inducible and mycoviral-regulated laccase3 (lac3) from the chestnut blight fungus Cryphonectria parasitica has recently been identified, but further characterization was hampered because of the precipitation of protein products by tannic acid supplementation. The present study investigated the heterologous expression of the functional laccase3 using a yeast Saccharomyce...
متن کاملA host factor involved in hypovirus symptom expression in the chestnut blight fungus, Cryphonectria parasitica.
The prototype hypovirus CHV1-EP713 causes virulence attenuation and severe suppression of asexual sporulation and pigmentation in its host, the chestnut blight fungus, Cryphonectria parasitica. We identified a factor associated with symptom induction in C. parasitica using a transformation of C. parasitica strain EP155 with a full-length cDNA clone from a mild mutant virus strain, Cys(72). This...
